Spore to Glass: The Brewers Turning Mold Into Their Most Powerful Ingredient
Mold has never exactly been a selling point. Walk into any brewery and you'll find a near-obsessive commitment to sanitation — every surface wiped, every vessel sterilized, every rogue microbe hunted down before it can wreck a batch. So when a handful of American craft brewers started deliberately inoculating their grain with a fuzzy white fungus, the industry noticed. Some were intrigued. Others thought these folks had completely lost the plot.
Turns out, they were just a few steps ahead.
What Is Koji, Exactly?
If you've eaten miso soup, soy sauce, sake, or a well-aged piece of Japanese charcuterie, you've already met Aspergillus oryzae — better known as koji. This controlled mold has been a cornerstone of East and Southeast Asian food culture for over a thousand years, used to break down starches and proteins in ways that produce deep, savory, umami-rich flavors. It's the metabolic engine behind an enormous chunk of fermented Asian cuisine.
What makes koji genuinely remarkable — and what caught brewers' attention — is its enzymatic firepower. The mold secretes a suite of enzymes, amylases and proteases chief among them, that essentially pre-digest whatever substrate they colonize. Starches get converted to sugars. Proteins get cleaved into amino acids. The result is a kind of flavor transformation that's difficult to replicate through any other means.
For brewers already obsessed with fermentation science, that enzymatic profile reads less like a contamination risk and more like a toolbox.
The Brewers Who Took the Leap
Dave Temkin at Almanac Beer Co. in the Bay Area was among the early adopters who started experimenting seriously with koji in an American brewing context. The premise was straightforward enough: grow koji on malted barley, then use that inoculated grain in the mash. What wasn't straightforward was everything that came after.
"The first few batches were genuinely disorienting," Temkin has said in interviews. "You're getting this incredibly smooth mouthfeel, these almost savory mid-palate notes — things that beer just doesn't normally do. We had to completely rethink how we were evaluating what we were making."
That disorientation is a recurring theme among koji brewers. The mold doesn't just add flavor notes the way a hop addition or a fruit adjunct would. It fundamentally rewires the fermentation chemistry, producing compounds — particularly a class of molecules called kojic acid and various amino-carbonyl reaction products — that create flavors described variously as mushroomy, meaty, caramel-adjacent, or simply "deep in a way that's hard to name."
In Portland, Oregon, Breakside Brewery has run several koji-forward experimental releases, pairing the mold-treated grain with koji-fermented rice additions to layer complexity. In Brooklyn, Interboro Spirits & Ales has explored koji in the context of hybrid beer-sake-style fermentations. Down in North Carolina, small-batch brewers operating out of taprooms and pilot systems are running koji trials that rarely make it beyond their own zip codes — but the results are circulating in brewing communities online, generating serious buzz.
Borrowing From Tradition Without Appropriating It
It's worth pausing here, because the conversation around koji in American craft brewing isn't just a technical one. There's a cultural dimension that thoughtful brewers are actively grappling with.
Koji is not some obscure ancient secret that Western brewers "discovered." It's a living, practiced tradition maintained by toji (sake master brewers) in Japan, by doenjang and ganjang producers in Korea, by makers of shaoxing wine in China, and by countless other practitioners across Asia. The knowledge is sophisticated, refined over generations, and deeply embedded in specific cultural contexts.
Some of the brewers doing the most interesting work with koji in the US are actively acknowledging that lineage — collaborating with Japanese-American food producers, consulting with sake brewers, and being transparent about where the technique comes from. Others are less careful. As with any instance of cultural borrowing in food and drink, the difference between appreciation and extraction often comes down to whether you're centering the tradition or just mining it for novelty.
The brewers worth paying attention to are the ones treating koji not as a trend to capitalize on, but as a fermentation philosophy to genuinely understand.
Beyond Koji: The Wider Mold Frontier
Koji gets most of the headlines, but it's not the only fungal culture making inroads in experimental brewing circles. Tempeh starter cultures — which use Rhizopus molds rather than Aspergillus — are being tested by a handful of brewers interested in their protein-transforming properties. Some mycology-minded fermenters are exploring the use of food-safe Monascus molds, which produce a striking red pigment along with a distinctive earthy flavor profile used traditionally in red yeast rice.
There's also growing interest in combining mold-based fermentation with wild yeast and bacteria cultures — layering koji's enzymatic pre-treatment with the tartness of lactobacillus or the barnyard funk of wild saisons. The results are predictably unpredictable, which is kind of the point.
Jordan Trigg, a homebrew-turned-professional brewer who now runs a small production operation in Asheville, NC, describes his approach as "mycological layering." He grows koji on malted wheat, uses it to build a base wort, then ferments with a mixed culture of his own wild-caught yeasts. The beers are genuinely difficult to categorize — not quite sour, not quite saison, not quite anything with an established name.
"I stopped trying to describe them in beer terms," Trigg says. "I just tell people it tastes like something they haven't had before. Which is true."
What It Actually Tastes Like
Fair question. The flavor experience of a well-made koji beer is legitimately hard to pin down if you're coming at it from a conventional beer background. The most common descriptors that come up among tasters: a silky, almost oily texture; a savory quality that sits somewhere between umami and caramel; a sweetness that doesn't read as sugary; and a long, complex finish that lingers in a way most beers don't.
Some koji beers lean more toward the savory end — almost broth-like, which sounds weird but works in certain contexts, particularly in lower-alcohol session formats. Others use the mold's enzymatic activity to build a base that amplifies fruit or hop character in unexpected ways, with the koji acting more as a flavor amplifier than a flavor itself.
The texture piece is consistently what surprises people most. Koji's protease activity breaks down proteins in ways that create a mouthfeel that beer drinkers don't have a ready vocabulary for. "Plush" comes up a lot. So does "coating."
The Road Ahead
Koji brewing in the US is still early-stage. There's no established playbook, no widely agreed-upon technique, and commercial examples remain genuinely rare outside of craft-focused taprooms and specialty bottle shops. But the trajectory feels meaningful. The brewers working in this space aren't chasing a trend — they're doing the kind of slow, iterative, curiosity-driven work that historically precedes real shifts in what American beer can be.
The mold, it turns out, isn't the enemy. It might be the next great collaborator.
